Detailed separation procedure

Mouse naive CD4
+
T cells are isolated by depletion of memory CD4
+
T cells and non-CD4
+
T cells. Single-cell suspensions of mouse spleen or lymph nodes are indirectly magnetically labeled with a cocktail of biotinylated monoclonal antibodies, as primary labeling reagent, and Anti-Biotin MicroBeads, as secondary labeling reagent. Simultaneously, CD44 MicroBeads are added to label memory T cells. No washing steps are required during magnetic labeling. The magnetically labeled non-target cells are depleted by retaining them on a MACS
®
Column in the magnetic field of a MACS Separator, while the unlabeled target cells pass through the column.
