As a control, CD184 (CXCR4) antibody staining was omitted and cells were measured in the same channels. Flow cytometry was performed with the MACSQuant® Analyzer. Cell debris, dead cells, and cell doublets were excluded from the analysis based on scatter signals and 4',6-diamidino-2-phenylindole (DAPI) fluorescence. No FcR Blocking Reagent was used. The recommended titers of respective antibodies from different suppliers were used.

To provide an indication on how an antibody performs after fixation of cells, in-house data on staining results before and after fixation with 3.7% formaldehyde using Miltenyi Biotec antibodies are provided. Different experimental settings may lead to different results.

Overview

The monoclonal antibody 12G5 recognizes human CD184, also known as CXCR4 or fusin, a 45 kDa seven transmembrane G-protein–linked chemokine receptor for SDF-1. CXCR4 is ubiquitously expressed on blood and tissue cells. It mediates chemotaxis in mature and progenitor blood cells and is important for B lympho- and myelopoiesis and cardiogenesis. It is reported to block CD4-independent HIV-2 infection and CD4-dependent infection by some T cell tropic isolates of HIV-1.
