Background information

The CD34 antigen is the most common marker for the isolation of hematopoietic stem and progenitor cells, but is also expressed on some lineage-committed cells. With the Diamond CD34 Isolation Kit, lineage-positive cells are removed in order to obtain pure, primitive CD34
+
HSCs.

Detailed separation procedure

The Diamond CD34 MicroBead Kit includes a lineage depletion followed by a CD34 isolation. In the first step, cells are labeled with a cocktail of biotin-conjugated antibodies against lineage-specific antigens. These cells are subsequently labeled with Anti-Biotin MicroBeads. Lineage-negative stem and progenitor cells are obtained by depletion of the magnetically labeled cells. Then CD34
+
cells are isolated from the lineage-negative stem and progenitor cells using CD34 MicroBeads. The resulting population obtained are CD34
+
Lin
stem cells.
