Background information

The StemMACS HSC-CFU Assay Kit offers a highly standardized method for analyzing hematopoietic stem and progenitor cells. The kit combines differentiation in methylcellulose-free cell culture with a standardized, flow cytometry-based read-out and eliminates the need for user-dependent, visual scoring under a microscope.
For the colony identification via flow cytometric readout when using the MACSQuant Analyzer, we provide an analysis excel sheet and a gating template. These analysis tools are optimized for the MACSQuantify Software version 2.11 and upwards. To receive these analysis tools, please contact your local technical support.

Technical data

Workflow using the StemMACS HSC-CFU Assay Kit.
HSCs, diluted in StemMACS HSC-CFU Assay Media, are plated in 96-well plates. After proliferation and differentiation for 14 days, the generated colonies are stained with the StemMACS HSC-CFU Assay Cocktail and analyzed by flow cytometry.
BFU-E: burst-forming unit-erythroid, CFU-G: colony-forming unit-granulocyte, CFU-M: CFU-macrophage, CFU-GM: CFU-granulocyte macrophage, CFU-GEMM: CFU-granulocyte erythrocyte macrophage megakaryocyte.
