This is basically a glorified blender that dissociates any tissue digested with collagenase, to extract infiltrating cells. It can be used for tumors, pancreases, kidneys, heart etc. Some solid tissues are difficult to dissociate by manual agitation, and this is where it comes handy. Plus, who wants to hurt their hands when you have a million tissues to process. We actually performed a yield test and found that tumors dissociated with this instrument gave 5 times higher numbers of infiltrating leukocytes than manually agitated samples.
Miltenyi Biotech's C-Tubes are designed to streamline tissue dissociation using the gentleMACS™ Dissociator, and are very helpful in doing so. By using set programs on the dissociator machine, you can select for specific cell types that you wish to isolate. I used these to dissociate tumor tissue in a cell line derivation procedure and received consistent, thoroughly positive results with my final cell lines.

Our lab focused on the clonal heterogeneity in various tissues. As part of our project, need to isolate single cells from various tissues ( liver, lung, brain) from mouse and tumors. The reason we choose MACS dissociator because it has pre-programme optimized for single cell preparations - Lung, brain and tumor.
Our lab digests large amounts of both human and animal tissue for single cell isolation. This machine very quickly dissociates many types of tissue that cannot digest in enzyme buffer alone. We use it for lung, spleen, intestine, and lymph nodes.
My lab studies CD8 T-cell mediated immunity in the genital mucosa and so we utilize this dissociator to break up evolutionarily tough tissues to free and suspend the cells inside.
