O4

cells were isolated from P6 mouse brain tissue using the Neural Tissue Dissociation Kit (P), the gentleMACS™ Dissociator, FcR Blocking Reagent, Anti-O4 MicroBeads, a MiniMACS™ Separator, and two MS Columns. Cells were fluorescently stained with Anti-O4-PE prior to separation. Cells were analyzed using the MACSQuant® Analyzer.

Detailed product information

Background information

During oligodendrocyte development, late A2B5
+
oligodendrocyte progenitors start to express O4. As the cells differentiate, A2B5 expression ceases, whereas O4 expression persists. More mature cells start to synthesize galactocerebroside, MBP, PLP, MOG, and O1. In the peripheral nervous system, O4 expression is found on Schwann cells. Dissociated mouse brain tissue derived from P3–7 CD1 mice contains approximately 5–10% O4
+
cells.
For the most efficient isolation of O4
+
cells, P3–7 (postnatal day 3–7) rodents are recommended.

Columns

For positive selection: MS, LS, or autoMACS
®
Columns. For depletion: LD or autoMACS Columns.
