Cells were incubated with an excess of purified unconjugated CD8b (REA222) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

As a control, CD8b antibody staining was omitted and cells were measured in the same channels. Flow cytometry was performed with the MACSQuant® Analyzer. Overview

Clone REA222 recognizes rat CD8b, a member of the immunoglobulin superfamily also known as T cell surface glycoprotein Lyt-3. CD8b is expressed on cytotoxic T cells, thymocytes, and macrophages as a heterodimer with CD8a. CD8 identifies cytotoxic/suppressor T cells that interact with MHC class I bearing targets and plays a role in the process of T cell mediated killing. Flow cytometric analysis showed that CD8 T cells from lymph nodes or spleen of normal rats almost exclusively express the α/β heterodimer, regardless of the T cell receptor isotype. In contrast, natural killer cells carry only CD8a chains.
Additional information: Clone REA222 displays negligible binding to Fc receptors.
