Cells were incubated with an excess of purified unconjugated TdT (19-3) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

Overview

The monoclonal antibody 19-3 recognizes the terminal deoxynucleotidyl transferase (TdT), a template independent 60 KDa DNA polymerase. TdT catalyzes addition of deoxynucleotides to the 3'-OH terminus of oligonucleotide primer segments. Expression of TdT is found in immature lymphocytes during early differentiation where it is involved in generating antigen receptor diversity by mediating Ig gene rearrangement.
