Specificity

Knockout validation
To ensure antibody specificity, the target gene is knocked out in a suitable cell line using the CRISPR/Cas9 system and the knockout is confirmed by sequencing of the target locus. The antibody is considered to bind specifically to the intended epitope if no antibody binding to the knockout cells can be detected. The antibody staining is controlled by fluorescence microscopy and/or flow cytometry.

Overview

Clone REAL339 is an antibody fragment derived from the full CD86 antibody molecule. It displays no binding to Fc receptors. The recombinantly engineered antibody fragments are multimerized to form the REAlease Complex to bind markers with high avidity.
CD86, also known as B7-2 or B70, is an 80 kDa molecule and a member of the immunoglobulin superfamily. Together with CD80 (B7-1) it belongs to the B7 family of co-stimulatory molecules. CD86 is expressed on activated B and T cells, dendritic cells, and monocytes/macrophages. The interaction of CD86 with its ligands CD28 and CD152 (CTLA- 4) plays a critical role in induction and regulation of immune responses, e.g., cross-talk between T and B cells, T cell costimulation, or immunoglobulin class-switching. Binding of CD86 to CD28 on T cells results in transduction of costimulatory signals for activation or proliferation of T cells, or cytokine production.
