The CD8a
+
T Cell Isolation Kit, mouse was developed for the untouched isolation of CD8a
+
T cells from single-cell suspensions from mouse spleen or lymph node. Purity has been improved with the release of this kit due to the addition of TCRγ/δ antibody to the depletion cocktail.
This kit uses the new fast mouse protocol and replaces the CD8a
+
T Cell Isolation Kit II (# 130-095-236).

T cells were isolated from this single-cell suspension using the CD8a+ T Cell Isolation Kit, an LS Column, and a MidiMACS™ Separator. Cells were fluorescently stained with the MC CD8a T Cell Cocktail (# 130-092-915) and analyzed by flow cytometry using the MACSQuant® Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.

Detailed product information

Background information

The kit is based on a cocktail of biotin-conjugated antibodies against CD4, CD11b, CD11c, CD19, CD45R (B220), CD49b (DX5), CD105, Anti-MHC-class II, Ter-119 and TCRγ/δ. The isolation of T cells is achieved by depletion of magnetically labeled cells.

Downstream applications

Untouched T cells are used for functional studies in which cross-linking of surface molecules should be excluded. Isolated T cells can also be further magnetically separated into T cell subsets.
The CD8a
+
T Cell Isolation Kit has been used for the following applications: For adoptive transfer experiments
1,2
, T cell proliferation assays
2,3
, Coculture with dendritic cells
2
and Cytokine analysis experiments.

CD8a+ T Cell Isolation Kit, mouse (130-104-075)

Our lab is interested in isolating CD8 T cells after harvesting spleens from mice. After isolation, we wanted to look at the CD8 T cell gene expression under varying conditions of glutamine concentration. The isolation was reliable at times but not clean other times.
