The sorting mechanism of the MACSQuant Tyto Cell Sorter. Coming from the input chamber, cells enter the microchip through a microchannel where they are interrogated by three lasers. Before entering the microchannel, potential cell aggregates are held back by a filter system, guaranteeing a smooth sorting process.
When a target cell (magenta) is identified, a magnetic pulse coming from the solenoid opens the microvalve, which then redirects the target cell into the positive collection chamber. In the default state, the valve is closed allowing non-selected cells (blue and orange) to flow through into the negative collection chamber.

In addition, the MACSQuant Tyto Cartridge is a single-use disposable that diminishes any risk of sample cross-contamination. Both positive and negative sorted fractions remain in the MACSQuant Tyto Cartridge after the actual sort, allowing full retrieval of all cells within the sample.

Background information

The MACSQuant Tyto Cartridge consists of three compartments that can each be accessed through individual, aseptic Luer-Locks from above: the input chamber, the positive collection chamber and the negative collection chamber.
Cells suspended in the specially formulated MACSQuant Tyto Running Buffer are loaded into the input chamber of the MACSQuant Tyto Cartridge. The input chamber contains an adjustable propeller, which keeps cells in suspension during the sort.
After loading the cartridge into the instrument, filtered air is applied to the input chamber through a pressure inlet. Driven by very low air pressure (~3 psi), fluorescently labeled cells flow from the input chamber, via a microchannel, through the microchip. Within the microchip, cells are interrogated by three lasers. Fluorescent and scatter light signatures are used to select target cells, which are then redirected by a sort valve located within the microchannel.
To redirect a target cell into the positive collection chamber, a magnetic field is applied to the microchip. This triggers the sorting valve to open and therefore change the flow of the target cell towards the positive collection chamber.
After a target cell has passed through, a silicon spring returns the valve into its original position.
Unlike during conventional droplet sorting procedures, the cells do not experience high pressure, they are not decompressed, and no charge is applied, resulting in great viability and functionality of cells.
