Background information

CD4
+
DCs coexpress CD11c, CD11b, MHC class II, CD40, CD80, CD86 and are negative for CD8 and CD205. They represent the major population of DCs in spleen and comprise only a small population in peripheral lymph nodes (56% and 4% of all CD11c
+
DCs, respectively). The isolation of CD4
+
DCs is performed in two steps.

Detailed separation procedure

First, DCs are enriched by depletion of T cells, NK cells, B cells, and granulocytes with a cocktail of biotin-conjugated antibodies and Anti-Biotin MicroBeads. Subsequently, CD4
+
DCs are positively selected from the enriched DC fraction with CD4 MicroBeads.

Dendritic Cell Isolation Kit are suitable to analyze antigen uptake, antigen processing, T cell activation or T cell tolerance induction, cross-priming of cytotoxic T cells, and T cell polarization by CD4
