Which phase contains basophils after Ficoll density gradient centrifugation? Is it possible to detect the basophils based on their forward scatter (FSC) or side scatter (SSC) in flow cytometry analysis?

Basophils are found in the interphase (PBMC fraction) of the Ficoll preparation. It is not possible to identify basophils by size and granularity, because their FSC and SSC properties are similar to lymphocytes.
