Which cell lines can be used as positive control for CD133 staining?

The Y79 or WERI-Rb1 cell lines can be used as a positive control for the fluorochrome-conjugated CD133 clones CD133/1 and CD 133/2. Alternative cell lines include NT-2 and Caco-2.

It is also possible to stain CD133+ cells in peripheral blood mononuclear cells (PBMCs). However, since the frequency of CD133+ cells is quite low, you must acquire a lot of cells or perform a pre-enrichment of the cells first using CD34 MicroBeads before staining them. Approximately 35­–75% of the CD34+ cells should be also CD133+.
