T cells from human PBMCs.

Background information

CLA (cutaneous lymphocyte-associated antigen) is an inducible carbohydrate modification of P-selectin glycoprotein ligand 1 (PSGL-1) that functions as a skin-homing receptor. CLA is found on a minor subset of human T cells that infiltrate the skin. The ligand of CLA, E-selectin, is expressed on vascular endothelium.
In peripheral blood, CLA is not only expressed on CD4
+
memory/effector T cells but also on subsets of CD8
+
T cells, B cells, NK cells, dendritic cells, and monocytes.

Detailed separation procedure

First, non-CD4
+
T cells are first magnetically labeled and depleted using a cocktail of biotin-conjugated antibodies and Anti-Biotin MicroBeads. Thereafter, the CD4
+
CLA
+
T cells are indirectly magnetically labeled with Anti-CLA-PE and Anti-PE MicroBeads to be isolated by positive selection.
