CD141 (BDCA-3) MicroBead Kit, human

The CD141 (BDCA-3) MicroBead Kit has been developed for the isolation of CD141 (BDCA-3)
cells (namely type-2 MDCs or MDC2) from PBMCs. CD141 (BDCA-3)
is expressed at high levels only on a minor subpopulation of myeloid dendritic cells (MDCs) in human blood (about 0.04 % of all PBMCs).

MDCs are CD11c

, CD1c (BDCA-1)
, CD4
, and do not express lineage markers such as CD3, CD14, CD16, CD19, CD20, and CD56. They express myeloid markers such as CD13 and CD33 and are monocytoid in appearance (fig. 2). In contrast to CD1c (BDCA-1)
MDCs, CD141 (BDCA-3)
MDCs do not express CD2 and Fc receptors such as CD32, CD64, and FceRI. Furthermore, CD141 (BDCA-3)
MDCs differ from CD1c (BDCA-1)
MDCs in terms of Toll-like receptor expression, cytokine production, and T helper cell polarization.
In order to discriminate the CD141 (BDCA-3)
from CD1c (BDCA-1)
MDCs, they have been designated type-2 MDCs (MDC2s).
The CD141 (BDCA-3) antigen is also expressed at a much lower level on CD1c (BDCA-1)
MDCs, plasmacytoid dendritic cells, monocytes, and granulocytes in blood.

MDCs were separated for their further phenotypic and functional characterization
, for gene clustering
, or for expression analyses of newly defined antigen EMR2 on different dendritic cell subsets
. The CD141 (BDCA-3) MicroBead Kit was also used for isolation of human CD141 (BDCA-3)
MDCs from mice engrafted with CD34
hematopoietic progenitors.
