stimulation and subsequent isolation of NY-ESO-1-specific CD4

T cells.

Background information

NY-ESO-1 is a cancer/testis antigen expressed in normal adult tissues solely in the testicular germ cells of normal adults and in various cancers, such as melanoma and breast, ovarian, prostate, lung and bladder cancer. The NY-ESO-1 protein is highly immunogenic and has become an attractive target for anticancer immunotherapy.

Applications

The
in vitro
stimulation of NY-ESO-1-specific CD4
+
and CD8
+
T cells with MACS GMP PepTivator NY-ESO-1 results in production of the effector cytokine IFN-gamma. The presence of IFN-gamma on the cell surface then permits the enrichment of NY-ESO-1-specific effector/memory T cells with the use of the CliniMACS Cytokine Capture System (IFN-gamma). Adoptive transfer of NY-ESO-1-specific T cells or vaccination with NY-ESO-1-pulsed dendritic cells may be an effective tool for the treatment of NY-ESO-1-expressing malignancies.
