Background information

The MAGE-A3 belongs to the melanoma antigen family and all members encode proteins with a high sequence similarity of up to 80% to each other. The expression of MAGE-A3 is restricted to the germ cells of testis but absent in somatic tissue except tumor cells.
In diagnostic application the protein is used for the identification of tumors like melanoma, non-small-lung cancer and hematologic malignancies. This makes MAGE-A3 to potent tumor-marker for immunotherapy.

Applications

The MACS GMP PepTivator MAGE-A3 is suitable for
in vitro
stimulation of CD4
+
and CD8
+
effector and memory T cells. The activated MAGE-A3-specific T cells secret IFN-gamma and can be enriched via the CliniMACS Cytokine Capture System (IFN-gamma).
Adoptive transfer of MAGE-A3-specific T cells or vaccination with MAGE-A3-pulsed dendritic cells may be an effective tool for the treatment of MAGE-A3-expressing malignancies.
