Background information

Tissue clearing refers to the process of rendering opaque tissues optically transparent. Working with transparent tissues is required for the imaging analysis of large biological systems in three dimensions (3D).
The MACS
®
Clearing Kit provides reagents for the immunostaining and clearing of different tissues. Briefly, samples are first fixed and permeabilized prior immunostaining. The duration of the immunostaining step is dependent on the tissue. After immunostaining, tissues are dehydrated using a series of ethanol dilutions and afterwards treated to render them transparent (cleared). Cleared tissues can be analyzed using the MACS
®
Imaging Solution in different microscopy systems.
Specific protocols using the MACS
®
Clearing Kit are available for different tissues, such as mouse brain hemispheres, cerebral organoids, and human tumors. The MACS
®
Clearing Kit is supplied with sufficient amounts to clear high number of tissues, such as for instance up to 20 mouse brain hemispheres.

Clearing Kit. Duration of immunostaining step is based on the use of directly conjugated antibodies.
*Organoids need to be embedded in a gel prior dehydration. This steps takes approximately 30min.
