Cells were incubated with an excess of purified unconjugated CD196 (CCR6) (REA277) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

Overview

Clone REA277 recognizes the CD196 antigen, a G protein–linked chemokine receptor also known as C-C chemokine receptor type 6 (CCR6). CD196 is preferentially expressed by B cells, immature dendritic cells, and subsets of T cells. The ligand of CD196 is macrophage inflammatory protein 3 alpha (MIP-3α) and binding subsequently transduces a signal by increasing the intracellular calcium ions level. CD196 has been shown to be important for B-lineage maturation and antigen-driven B cell differentiation, and it may regulate the migration and recruitment of dendritic and T cells during inflammatory and immunological responses. Targeting CD196 or its ligand may be a promising therapeutic strategy to alleviate atherosclerosis.
Additional information: Clone REA277 displays negligible binding to Fc receptors.
