  A single-cell suspension was prepared using the Epidermis Dissociation Kit, mouse. Cells were fluorescently stained with CD326 (EpCAM) antibodies and analyzed by flow cytometry using the MACSQuant ® Analyzer. CD11b positive cells were pre-gated for the analysis. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence or 4',6-diamidino-2-phenylindole (DAPI) fluorescence, as in the case of tandem conjugates.

  HM1 stem cell line cultured on feeder cells were fluorescently stained with CD326 (EpCAM) antibodies as well as with Anti-Feeder antibodies and analyzed by flow cytometry using the MACSQuant Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.

  Immunofluorescent microscopic images of fixed mouse embryonic stem cells (HM1) cultured on feeder cells: cells were fixed in PFA and stained with CD326 (EpCAM)-Biotin overnight at 4 °C. Detection of CD326 (EpCAM)-Biotin was performed with a secondary antibody (red).

  Cells were additionally stained with Anti-Oct4 antibody overnight. Detection of the Anti-Oct4 antibody was performed with a secondary antibody (green). Cell nuclei of stem cells and feeder cells were detected with DAPI (blue).
