We use CD34 as a marker gene in our retroviral vector used for the transduction of T cells. We use CD34 because it is not expressed on T cells and we can use reagents to purify our transduced populations and visualize them via flow using CD34 antibody staining. We use these CD34 immuno magnetic beads to purify our T cell populations to only include transduced T cells after the transduction process.
