Background information

Gp100/Pmel 17 belongs to the melanoma-associated antigens of the group of differentiation antigens. Other members of this group are, for example, tyrosinase and Melan-A/MART-1. Gp100/Pmel 17 is expressed on normal melanocytes, but is also present in melanoma. CD4
+
and CD8
+
T lymphocytes recognizing gp100/Pmel 17 have been identified in melanoma patients.

Downstream applications

The
in vitro
stimulation of gp100/Pmel 17 CD4
+
and CD8
+
T cells with PepTivator gp100/Pmel 17 causes secretion of effector cytokines and upregulation of activation markers, which then allow the detection and isolation of gp100/Pmel 17–specific T cells.
  • Detection and analysis of gp100/Pmel 17–specific CD4+ and CD8+ effector/memory T cells in PBMCs by MACS® Cytokine Secretion Assays, intracellular cytokine staining, or other technologies
  • Isolation of viable gp100/Pmel 17–specific CD4+ T cells with the CD154 MicroBead Kit, or of CD4+ and CD8+ T cells using the CD137 MicroBead Kit or MACS Cytokine Secretion Assay – Cell Enrichment and Detection Kits. Subsequently, cells can be expanded for generation of T cell lines.
  • Generation of gp100/Pmel 17–specific CD4+ and CD8+ effector/memory T cells for research on immunotherapy and vaccination
