Cells were incubated with an excess of purified unconjugated PAX-5 (BSAP) (REA140) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

Overview

Clone REA140 recognizes PAX-5, which belongs to the paired box (PAX) gene family of transcription factors. PAX-5 is the only mammalian PAX transcription factor, which is expressed in hematopoietic cells. Within the hematopoietic cells, expression of PAX-5 is limited to cells of the B-lymphocyte lineage and it exerts transcriptional control of B cell commitment, development, and tumorigenesis. PAX-5 contains DNA binding conserved ‘paired’ domain, which consists of an N- and a C-terminal subdomain. Each subregion contains a homeodomain-like helix-turn-helix motif, which binds to a half-site of the PAX-5 recognition sequence. Depending upon the interaction partners, PAX-5 serves either as an activator or repressor of transcription. For instance, Groucho protein family members, by binding to a conserved octapeptide motif of PAX-5, enable PAX-5 to serve as a repressor.
Additional information: Clone REA140 displays negligible binding to Fc receptors.
