T cells were isolated from a single-cell suspension from mouse spleen using the Naive CD8a
+
T Cell Isolation Kit, an LS Column, and a MidiMACS™ Separator. Cells were fluorescently stained with CD3ε-APC (# 130-092-977), CD8a-VioBlue (# 130-094-360), CD44-FITC (# 130-096-842), and CD62L-PE (# 130-091-756) and analyzed by flow cytometry using the MACSQuant
®
Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.

Overview

The Naive CD8a
+
T Cell Isolation Kit was developed for the isolation of untouched naive CD8a
+
T cells from mouse spleen and lymph node cells.

Detailed product information

Detailed separation procedure

For magnetic depletion of memory CD8
+
T cells and non-CD8
+
T cells, single cell suspension is incubated with a cocktail of biotinylated CD45R, CD4, CD11b, CD11c, CD19, CD44, CD49b, CD105, Ter-119, MHC II, and anti-TCRγ/δ antibodies. These cells are subsequently magnetically labeled with Anti-Biotin MicroBeads for depletion.

Downstream applications

Naive CD8a
+
T cells can be used for further
in vitro
analysis of T cell differentiation or adoptive transfer experiments.

Columns

LS, XS, or autoMACS
®
Columns.
