Knockout validation
To ensure antibody specificity, the target gene is knocked out in a suitable cell line using the CRISPR/Cas9 system and the knockout is confirmed by sequencing of the target locus. The antibody is considered to bind specifically to the intended epitope if no antibody binding to the knockout cells can be detected. The antibody staining is controlled by fluorescence microscopy and/or flow cytometry.

Overview

Clone REA751 recognizes the human claudin-3 antigen, an integral membrane protein which is a component of the claudin family of epithelial tight junction proteins. Claudin-3 is involved in tight junction-specific obliteration of the intercellular space. It is a low-affinity receptor for
Clostridium perfringens
enterotoxinin (CPE). Binding of CPE to claudin-3 induces epithelial cell lysis.
Additional information: Clone REA751 displays negligible binding to Fc receptors.
