Description:

Human myeloma U266 cells were stained with CD70 antibodies and analyzed by flow cytometry using the MACSQuant^® Analyzer. The specificity of the conjugated antibodies was confirmed by blocking the binding to the ligand, using pure unconjugated antibodies (left peak). Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.

Specificity

Knockout validation

To ensure antibody specificity, the target gene is knocked out in a suitable cell line using the CRISPR/Cas9 system and the knockout is confirmed by sequencing of the target locus. The antibody is considered to bind specifically to the intended epitope if no antibody binding to the knockout cells can be detected. The antibody staining is controlled by fluorescence microscopy and/or flow cytometry.

Overview

Clone REA292 recognizes the human CD70 antigen, a 50 kDa type II transmembrane glycoprotein also known as CD27 ligand (CD27L) or tumor necrosis factor ligand superfamily member 7 (TNFSF7). CD70 is a member of the TNF superfamily and is expressed by activated mouse T, B, and NK lymphocytes, activated plasmacytoid dendritic cells (pDCs), chronic B cell lymphocytic leukemia, and large B cell lymphomas. It interacts with CD27, its ligand on T cells, in order to promote T-B cell cross-stimulation and co-stimulates B cell proliferation and immunoglobulin production. It also plays a role in the pDC-induced B cell differentiation.

Additional information: Clone REA292 displays negligible binding to Fc receptors.
