Cells were incubated with an excess of purified unconjugated CD191 (CCR1) (REA158) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

To provide an indication on how an antibody performs after fixation of cells, in-house data on staining results before and after fixation with 3.7% formaldehyde using Miltenyi Biotec antibodies are provided. Different experimental settings may lead to different results.

Overview

Clone REA158 recognizes CCR1, a chemokine receptor that binds to CC subgroup of chemokines as ligands. Chemokines are small chemoattractant protein molcules and are subdivided into families CC, CXC, or CX3C.Chemokines exert their action via binding to corresponding chemokine receptors, which are G protein–coupled receptors and share several structural features. CCR1 binds chemokines auch as CCL3, CCL5, CCL7, and CCL14 as ligands. Expression of CCR1 is found on macrophages, natural killer cells, T cells, DC, monocytes, basophils, and eosinophils, where regulated CCR1 expression can be achieved by molecules such as IL2 and TGF-β2. CCR1-ligand interaction plays role in leukocyte migration, cell differentiation/activation and immune response.
Additional information: Clone REA158 displays negligible binding to Fc receptors.
