Cells were incubated with an excess of purified unconjugated Cardiac Troponin T (REA400) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

Overview

Clone REA400 recognizes the human, mouse, and rat cardiac troponin T antigen, a protein which is exclusively expressed at high concentrations in cardiac muscle and has emerged as the preferred biomarker in the universal definition of myocardial infarction. Cardiac troponin T is the tropomyosin-binding subunit of the troponin complex, which is located on the thin filament of striated muscles and regulates muscle contraction in response to alterations in intracellular calcium ion concentration. Mutations in the cardiac troponin T gene have been associated with familial hypertrophic cardiomyopathy as well as with restrictive and dilated cardiomyopathy. Transcripts for this gene undergo alternative splicing that results in many tissue-specific isoforms. Mutations of this gene may be associated with mild or absent hypertrophy and predominant restrictive disease, with a high risk of sudden cardiac death. Pluripotent stem cells (PSCs) differentiate into cardiomyocytes positive for cardiac troponin T and showing spontaneous rhythmic contractions. Therefore, PSC-derived cardiomyocytes have emerged as a potential source for cardiac regeneration after myocardial infarction.
Additional information: Clone REA400 displays negligible binding to Fc receptors.
