Background information
FFPE tissue sections from various tissues and species, for example, human tumor, skin, or lymph node samples, can be dissociated into single-cell suspensions by combining mechanical dissociation with enzymatic degradation of the extracellular matrix. The FFPE tissue sections are deparaffinized in this process. After a defixation step, samples are dissociated using the kit components and a gentleMACS™ Dissociator with Heaters. Larger particles are removed subsequently using MACS
® SmartStrainers.
This kit is specifically designed to achieve high single-cell yields while obtaining cytokeratin and vimentin epitopes relevant for carcinoma cell analysis. Single-cell suspensions are indicated for subsequent isolation of carcinoma cells, flow cytometric analysis, or molecular analysis.
This kit also allows the dissociation of FFPE tissue for single-cell sequencing using the 10x Genomics Chromium Single Cell Fixed RNA Profiling Assay. For this application you must follow the 10x Genomics demonstrated protocol is provided at this
