StraightFrom® LRSC MicroBeads allow for magnetic cell separation directly from Leukoreduction System Chambers (LRSCs). Leukocyte subsets such as CD4+, CD8+, CD14+, CD19+, or CD56+ cells can be purified using using either MultiMACS™ Instruments or manual separators. Pre-enrichment of white blood cells, e.g. by density gradient centrifugation, is not required. The purified cells are well-suited for further flow cytometric analyses, molecular biology applications, and functional studies.

LRSCs arise after routine donor plateletpheresis procedures, during which unwanted leukocytes are filtered out into the LRSC, also known as buffy cone. As LRSCs contain highly concentrated leukocytes in a relatively small volume, they are an ideal source for leukocytes.

With StraightFrom LRSC MicroBead Kits cells can be directly isolated from an entire or partial LRSC. Together with LS columns (included in the kit) and magnetic separators such as the QuadroMACS™ Separator, leukocytes can be manually enriched. First, the MicroBeads are added directly to the blood sample without any prior cell counting or PBMC isolation. Leukocytes can then be isolated straightforward within minutes without the need for density gradient centrifugation.

The MultiMACS Cell24 Separator Plus is ideally suited to process an entire LRSC in a single run using StraightFrom LRSC MicroBead Kits. 

The 30-minute isolation procedure results in cell populations with high purities and recovery rates. With 12 LS Columns (included in the kit), the instrument allows for time saving processing of large sample volumes, while providing reliability and reproducibility in day-to-day cell separation. 

For large-scale cell separations, the MultiMACS X is the instrument of choice.

CD8+ cells can be isolated straight from a LRSC sample using the StraightFrom LRSC CD8 MicroBead Kit, human and the MultiMACS™ Cell24 Separator Plus.

Separation of a LRSC sample using the StraightFrom® LRSC CD8 MicroBead Kit, human and the MultiMACS™ Cell24 Separator Plus with the Single-Column Adapter and LS Columns. Cells were fluorescently stained with CD3-APC, CD8-PE, as well as CD45-VioBlue® and analyzed by flow cytometry using the MACSQuant® Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.
