Cells were incubated with an excess of purified unconjugated CD63 (REA1055) antibody followed by staining with fluorochrome-conjugated antibodies of other known clones against the same marker. Based on the fluorescence signal obtained, the clones were identified as recognizing completely overlapping (++), partially overlapping (+), or completely different epitopes (-) of the marker.

To provide an indication on how an antibody performs after fixation of cells, in-house data on staining results before and after fixation with 3.7% formaldehyde using Miltenyi Biotec antibodies are provided. Different experimental settings may lead to different results.

Overview

Clone REA1055 recognizes the human CD63 antigen, a type III glycoprotein belonging to the tetraspanin family of proteins, which are characterized by four membrane spanning domains. Tetraspanins via various homotypic and heterotypic interactions regulate the spatial location and trafficking of associated transmembrane partners, eventually regulating and co-ordinating various signaling pathways. CD63, like other tetraspanins, is present in the plasma membrane and membrane of various subcellular compartments such as late endosomes, lysososmes, cytotoxic granules of T lymphocytes, the crystalloid granules of eosinophils, the secretory granules of basophils, and mast cells. Interaction partners of CD63 include MHC class II molecules in immature dendritic cells and B lymphocytes, FcεRI in basophils and mast cells, etc.
Additional information: Clone REA1055 displays negligible binding to Fc receptors.
