Overview Plasmacytoid Dendritic Cell Isolation Kit II は、リンパ組織の単細胞懸濁液から非標識 plasmacytoid dendritic cells (PDCs) を分離するために開発されました。 Details Background information 分離したばかりのPDCsは、MHC class IIや共刺激分子の発現が低く、ヘルパーＴ細胞の刺激能が弱いです。特定の非メチル化CpGモチーフ (CpG-DNA) を有する細菌DNAによる刺激、あるいはウイルス感染により、PDCsは大量のⅠ型 IFN を産生し、抗原提示能を獲得します1,2。PDCsはmPDCA-1 (murine plasmacytoid dendritic cell antigen-1) を特異的に発現しているため、リンパ組織又は非リンパ組織中のPDCsをこの抗体によって直接同定することが出来ます3。最近、mPDCA-1は、bone marrow stromal antigen 2 (BST2) と同定されました4。 Detailed separation procedure Plasmacytoid Dendritic Cell Isolation Kit II では、PDCs以外の細胞を、ビオチン標識モノクローナル抗体カクテルと Anti-Biotin MicroBeadsで標識し、MACS Separatorの磁場内に置いたMACS® Columnに保持させることによって除去します。一方、カラムを素通りする非標識PDCsは、高純度に濃縮されます。 Downstream applications Plasmacytoid Dendritic Cell Isolation Kit II を用いて分離した PDCsは、PDCsによる抗原の取り込みやプロセシング、Ｔ細胞の活性化や寛容の誘導、細胞傷害性Ｔ細胞のクロスプライミング、ヘルパーＴ細胞のpolarizationの分析に適しています。 Columns MS, LS 又は autoMACS®Columns		Figure 1 PDCs were isolated from a mouse spleen cell suspension by using the Plasmacytoid Dendritic Cell Isolation Kit II, an LS Column, and a MidiMACS™ Separator.
