CD154 Detection Cocktails (mouse)
Overview CD154 Detection Cocktail は、活性化した抗原特異的 CD4+ T 細胞を簡単に同定できるように開発されました。細胞を in vitro で適当な抗原により刺激すると、CD154 の発現が誘導されます。このとき同時に CD154 Detection Cocktail を加えてインキュベーションします。 蛍光色素標識された CD154 抗体のほか、CD154 Detection Cocktail には、CD154 の細胞表面における発現のダウンレギュレーションを防ぐための CD40 ブロッキング抗体、および共刺激シグナルを入れるための CD28 抗体が含まれています。 Details Background information CD154 は、CD40L、gp39、T-BAM、TRAPとしても知られており、活性化した CD4+ T 細胞上で一過的にアップレギュレートされます。CD154 は、T 細胞と抗原提示細胞の相互作用において、CD40 との結合を介して共刺激分子として重要な役割を担っています。活性化後数時間内の一過性の発現のため、活性化した抗原特異的 CD4+ T細胞の信頼できるマーカーとしてCD154 を用いることが出来ます1,2。 Clone Isotype CD28 Hamster IgG2 CD40 Rat IgG2a CD154 Hamster IgG3		Figure 1 Mouse splenocytes were incubated with or without SEB for 6 hours in the presence of the CD154 Detection Cocktail (PE) or the CD154 Detection Cocktail (APC). IL-2 was detected using an IL-2 Secretion Assay – Detection Kit. Cells were stained with CD4-FITC and CD45R (B220)-PerCP™ and analyzed by flow cytometry. Lymphocytes were gated based on scatter properties. Dead cells and CD45R (B220)+ cells were excluded from the analysis.
