Description Treg Suppression Inspector は、ヒト CD4+CD25+ regulatory T (Treg) 細胞の機能的試験である in vitro 抑制アッセイのために開発されました。 Treg 細胞は、T 細胞のサブセットであり、自己抗原あるいは外来抗原に対する有害な免疫反応を抑制する能力をもちます。この Treg 細胞の機能は、いわゆる”抑制アッセイ”によって解析することができます。ポリクローナルな刺激、すなわち Treg Suppression Inspector の存在下で、レスポンダーである T 細胞と Treg 細胞とを、種々の存在比で共培養し、 T 細胞の増殖反応を測定します。 Treg Suppression Inspector は、Treg 抑制アッセイに最適化した T 細胞刺激試薬です。試薬は、Anti-Biotin MACSiBead™ Particles に、ビオチン化した CD2, CD3, CD28 抗体を結合したものです。 レスポンダーである T 細胞 (CD4+CD25- あるいは CD4+ T 細胞) のみを Treg Suppression Inspector 存在下で培養すると増殖が誘導されるのに対し、Treg 細胞のみを同様に培養した場合には有意な増殖は認められません。Treg 細胞とレスポンダー T 細胞を共培養すると、Treg 細胞の抑制機能により、レスポンダー T 細胞の増殖が減弱します。 Applications Treg Suppression Inspector は、 ヒト CD4+CD25+ Treg 細胞の in vitro 抑制アッセイによる機能解析のために開発されました。		Figure 1 CD4+CD25+ regulatory T cells, isolated with the CD4+CD25+ Regulatory T Cell Isolation Kit, were cocultured with CD4+CD25- responder T cells in different ratios. For T cell stimulation, the Treg Suppression Inspector was added to the culture. As controls, CD4+CD25+ Treg cells and CD4+CD25- responder T cells alone were cultured without any stimulus.
