Anti-mPDCA-1抗体 (マウス)

Overview

Anti-mPDCA-1 抗体は、フローサイトメトリーや蛍光顕微鏡法、免疫組織染色法においてマウス PDCs を単一カラーで直接検出することができます。この抗体は、組織より調製した細胞懸濁液から直接分離したPDCsや、骨髄細胞培養から in vitro で誘導してきた PDCsの解析に有用です。またin vivo での PDCs のディプリーションにも使用できます。さらに、Anti-mPDCA-1抗体は、感染や自己免疫疾患モデルマウスにおけるPDCsの組織での分布や存在頻度、リンパ組織内での特異的な局在、炎症部位への遊走の研究に使用できます。 Plasmacytoid Dendritic Cell Isolation Kit II, Anti-mPDCA-1 MicroBeads または CD11c MicroBeadsを用いて分離したPDCsの評価にも、この抗体の使用をお勧めします。

Background information
Clone JF05-1C2.4.1はMouse plasmacytoid dendritic cell antigen-1 (mPDCA-1) を認識します。mPDCA-1はマウス plasmacytoid dendritic cells (PDCs) に特異的に発現しています。PDCs は、CD11c⁺ 樹状細胞のサブセットで、すべてのリンパ組織、末梢血、そして一部の非リンパ組織において低頻度で見られます。
マウス脾臓、骨髄、リンパ節では、mPDCA-1 は CD11c^low, Ly-6C^high, CD45R (B220)⁺, MHC Class II^low, CD8a^low/-, CD80^low/-, CD86^-, CD40^-, CD11b^-, CD90^-, CD49b (DX5)^-, CD3^-, CD19^-、すなわちマウス PDCs の表現型をもつ細胞にのみ特異的に発現しています。脾臓細胞の蛍光多重染色によると、すべての CD11c^low Ly6C^high CD45R (B220)⁺ PDCs は mPDCA-1⁺ であり、それゆえ mPDCA-1 の発現は PDCs に特異的であると言えます。

Identification of CD11c⁺ Ly-6C⁺ CD45R (B220)⁺ PDCs with Anti-mPDCA-1.
Splenocytes isolated from BALB/c mice were analyzed by multi-color fluorescent staining with CD11c, Anti-Ly-6C, CD45R (B220), and Anti-mPDCA-1. Cells expressing Ly-6C^high CD45R (B220)^high (A) are also positive for CD11c and mPDCA-1 (B).
Note that all CD11c^low Ly-6C^high CD45R (B220)⁺ PDCs are mPDCA-1⁺ and that mPDCA-1 expression is restricted to PDCs only.

Direct identification of PDCs with Anti-mPDCA-1-FITC. Splenocytes were stained with Anti-PDCA-1-FITC and Anti-Ly-6C-PE. Note that staining with Anti-mPDCA-1-FITC alone is sufficient for direct detection of murine PDCs.
