Overview Annexin V MicroBead Kitは、Annexin Vに結合する能力に基いて細胞をポジティブセレクション又はディプリーションするために開発されました。Annexin V MicroBead Kitには、Annexin V MicroBeadsとAnnexin V Binding Bufferが含まれます。 Details Background information ほとんどの正常な生きた真核細胞では、マイナス荷電したリン脂質であるホスファチジルセリン(PS) は、細胞膜脂質２重層のcytosol側の層に存在しています。このPS の内層から外層への再分布は、アポトーシスの早い時期に広く観察される現象です。一方、ネクローシスでは、膜が壊れることによってPSへのアクセスが可能になります。ネクローシスとアポトーシス以外にも、活性化された血小板、鎌状赤血球貧血のようなある種の細胞異常1、赤血球の老化、マスト細胞の脱顆粒、さらにB細胞分化のある段階においても PS へのアクセスが可能になります。PS 露出はまた、マクロファージによる細胞の認識と除去も引き起こします。 Annexin Vは 35 kDa のリン脂質結合タンパク質であり、マクロファージやその他の貪食細胞の主要な細胞膜成分です。 Applications Annexin V MicroBead Kitは、鎌状赤血球やアポトーシス細胞など、PSの露出した細胞の分離に使用されました1-3。またAnnexin V MicroBead Kitは、好中球の生存を調節する細胞内シグナル伝達を研究するため、アポトーシス及びnon-アポトーシス好中球を分離するのに使用されました4。異なるcytosolic phospholipase A2sのアポトーシスにおける役割についての研究において、PS 露出の高感度検出にもAnnexin V MicroBead Kitが使用されました5。 Columns MS, LS 又は autoMACS® Columns		Figure 1 Isolation of PS-exposing cells using the Annexin V MicroBead Kit.
