Description CD4+CD25+CD127dim/– regulatory T細胞の分離は、ビオチン化抗体カクテルとAnti-Biotin MicroBeadsによるnon-CD4+ とCD127high 細胞のディプリーション、さらに引き続いてCD25 MicroBeadsⅡによるCD4+CD25+CD127dim/- 細胞のポジティブセレクションによって行います。 CD127 は IL-7 レセプターの α 鎖であり、ほとんどの成熟 T細胞に発現していて、その分化や増殖に重要な役割を担っています。しかしCD127 の発現は、ヒト制御性T 細胞ではダウンレギュレートされており、これは FoxP3 とは逆のパターンの発現となります。このため、ヒト制御性T細胞と活性化T細胞とを区別するためのマーカーの一つとして、CD127 を用いることが出来ます。 Applications ヒトPBMCからCD4+CD25+CD127dim/– regulatory T細胞を分離して、表現型や機能の解析を行えます。 Columns non-CD4+ 細胞とCD127high 細胞のディプリーション: LD 又は autoMACS™ Columns CD25+ 細胞のポジティブセレクション: MS 又は autoMACS Columns Disclaimer CD4+CD25+CD127dim/– regulatory T Cell アイソレーションキット（ヒト）（#130-093-337）の後継品です。		Figure 1 CD4+CD25+CD127dim/– regulatory T cells were isolated from human PBMCs by using the CD4+CD25+CD127dim/– Regulatory T Cell Isolation Kit II, an LD and two MS Columns, a MidiMACS™ Separator and a MiniMACS™ Separator. The cells were fluorescently stained with CD4-FITC, CD25-APC, and CD127-PE, or CD4-FITC, CD127-PE, and Anti-FoxP3-APC and analyzed by flow cytometry using the MACSQuant™ Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and PI fluorescence.
