Blood Dendritic Cell Isolation Kit II
Description Blood Dendritic Cell Isolation Kit II は、ヒト末梢血単核細胞（PBMCs）から plasmacytoid および myeloid dendritic cells の両方を合わせて分離するための磁気標識システムです。分離は２ステップの MACS® Separation で行います。まず、CD19 および CD14 MicroBeads のカクテルを使用して、B細胞と単球を磁気標識しディプリーションします。その後、磁気標識されていないネガティブフラクション中の濃縮樹状細胞を、樹状細胞マーカー CD304 (BDCA-4/ Neuropiline-1), CD141 (BDCA-3), CD1c (BDCA-1) に対する抗体カクテルを使用して磁気標識し、さらに濃縮します。高純度の濃縮細胞フラクションは、 plasmacytoid dendritic cells, CD1c (BDCA-1)+ type-1 myeloid dendritic cells (MDC1s), CD1c (BDCA-1)- CD141 (BDCA-3)bright type-2 myeloid dendritic cells (MDC2s) からなります。B細胞の一部は CD1c (BDCA-1) を発現し、単球は CD141 (BDCA-3) を弱く発現しているので、B細胞と単球を予めディプリーションします。 キットには、FcR Blocking Reagent, Non-Dendritic Cell Depletion Cocktail, ならびに Dendritic Cell Enrichment Cocktail が含まれています。Blood Dendritic Cell Isolation Kit II を使用した MACS® Separations の評価には、解析に必要なすべての抗体と isotype control が含まれている Blood Dendritic Cell Enumeration Kitをお勧めします。 Applications MACS® Technology で分離された樹状細胞は、例えば in vitro での primary T 細胞反応の誘導などに適しています。 Columns ステップ1の磁気分離（ディプリーション）には LD, CS, D, 又は autoMACS™ Columnsステップ2の磁気分離（ポジティブセレクション）にはMS, LS, 又は autoMACS Columns		Figure 1 Working scheme for the isolation of dendritic cells from PBMCs using the Blood Dendritic Cell Isolation Kit II.
