Toll-like receptor (TLR) 7 and 8 are involved in the response to viral infection. They recognize single stranded RNAs as their natural ligand and also small synthetic molecules.

We provide high quality ORNs for activation of either TLR7 only, TLR8 only, or TLR7 and TLR8 simultaneously, together with the respective controls.  All ORNs come with a transfection reagent for optimal performance. ORN R-2176-dT, exclusively available from Miltenyi Biotec, can also be used without transfection reagent.

Which cell type do you want to stimulate? Choose among our portfolio of TLR7, TLR8 or TLR7/8 agonists. All of our ORNs come with the appropriate transfection reagent (DOTAP-Cl) and protocol to enable safe, easy, and fast experimental set-up.

ORN R-2176-dT, a TLR7/8 agonist, can also be used without prior complexation to DOTAP-Cl, further simplifying cell activation.

In human cells, TLR7 Is predominantly expressed in plasmacytoid dendritic cells (pDCs) and B cells, whereas in mice, TLR7 is also expressed on myeloid dendritic cells (mDCs) and monocytes.

TLR7 agonist ORN R-2336 activates exclusively human and murine TLR7. This ORN induces, e.g., TLR7 specific IFN-α from human PBMC.

To enable uptake and protect them from degradation, ORNs need to be formulated with cationic lipids.  The recommended transfection reagent is DOTAP-Cl (1,2-Dioleoyloxy-3-trimethylammonium propane chloride).

In humans, TLR8 is mainly expressed on cells of the myeloid lineage, such as monocytes, macrophages, and myeloid dendritic cells (mDCs). Murine TLR 8 is functionally impaired. Thus, TLR8 agonists activate specifically human TLR8 - these ORNs are not active in the murine system. ORN R-0002 induces cytokines like IFN-γ, IL-12, and TNF-α, but not IFN-α production.

TLR7 and TLR8 are expressed on immune cells with a distinct expression pattern. In human cells, pDCs and B cells express predominantly TLR7 whereas TLR8 is mainly expressed on cells of the myeloid lineage. In mice, TLR7 is also expressed on mDCs and monocytes whereas murine TLR 8 is functionally impaired.

TLR7/8 agonists ORN R-0006, RNA 40, or ORN R-2176-dT, activate both human TLRs, 7 and 8, as well as murine TLR7 and induce a broad spectrum of cytokines, e.g. IFN-α, IFN-γ, IL-12 and TNF-α.
