Background information Interleukin-5 (IL-5) is predominantly secreted by CD4+ T cells. It promotes growth and activation of eosinophils, is involved in a range of allergic reactions, and mediates immune reactions against parasites. IL-5 also acts on other cell types such as B cells.
Downstream applications The Mouse IL-5 Secretion Assays can be used to detect and isolate murine antigen-specific, IL-5-secreting T cells after stimulation with antigen (fig. 1). The assays allow studies on TH2 development and commitment as well as on the mechanism of IL-5 expression. |

BALB/c mice were immunized and the spleen cells restimulated with keyhole limpet hemocyanin (KLH) as described for the Mouse IFN-γ Secretion Assay. The responding cells were stained and enriched according to secretion of IL-5 using the Mouse IL-5 Secretion Assay - Cell Enrichment and Detection Kit. In the stimulated sample, 233 IL-5-secreting CD4+ T cells were enriched per 106 CD4+ T cells using MS Columns and a MiniMACS™ Separator. In the unstimulated control sample, only six IL-5-secreting CD4+ T cells were detected per 106 CD4+ T cells after enrichment. |
