| Description |
The Stemgent™ Alkaline Phosphatase (AP) Staining Kit is a specific and sensitive tool for phenotypic assessment of ES cell differentiation or iPS cell generation from somatic cells. The undifferentiated state of ES cells and iPS cells can be characterized by a high level of AP expression1-4, which, along with the expression of surface markers including stage-specific embryonic antigens (SSEA-1, -3 and -4), TRA antigens (TRA-1-60, TRA-1-81), and the transcription factor Oct4, indicates a subset of cells with self-renewal potential.
AP is a hydrolase enzyme responsible for dephosphorylating molecules such as nucleotides, proteins, and alkaloids under alkaline conditions. When fixed ES cells or iPS cells are stained with AP, undifferentiated cells appear red or purple, whereas differentiated cells appear colorless. This feature allows for a simple, sensitive, and rapid assay for the visualization of undifferentiated pluripotent cells and colonies. |
