TCRγ/δ+ T Cell Isolation Kit
Description
The TCRγ/δ+ T Cell Isolation Kit has been developed for the isolation of untouched mouse TCRγ/δ+ T cells from single-cell suspensions of spleen, lymph nodes, or thymus.
Details
Background information
T cells bearing γ/δ TCRs are a phenotypically and functionally conserved lineage of T cells which often constitutes the major T cell population in epithelial-rich tissues, e.g. intestine, skin, lung, and reproductive tract. In blood and lymphoid tissues they are found at low frequency. TCRγ/δ+ T cells have been shown to be involved in immune surveillance against tumors, wound healing, as well as pro- and anti-inflammatory responses against pathogens.1
The TCRγ/δ+ T Cell Isolation Kit contains CD11b and CD45R (B220) MicroBeads for depletion of CD11b+ and CD45R (B220)+ cells, as well as Anti-TCRγ/δ-Biotin and Anti-Biotin MicroBeads for positive selection of TCRγ/δ+ T cells.

Downstream applications
TCRγ/δ+ T cells isolated by using the TCRγ/δ+ T Cell Isolation Kit are suitable for phenotypic and functional analysis, adoptive transfer, and coculture experiments.
Columns
For the first magnetic separation (depletion): LD or autoMACS Columns; for the second magnetic separation (positive selection): MS or autoMACS Columns.

Figure 1
TCRγ/δ+ T cells were isolated from single cell suspensions of mouse spleen (A-C), lymph node (D), and thymus (E) by using the TCRγ/δ+ T Cell Isolation Kit, an LD and two MS Columns, a MidiMACS™ and a MiniMACS™ Separator.

B: Pre-enriched splenic TCRγ/δ+ T cells after depletion of CD11b+ and CD45R (B220)+ cells
C: TCRγ/δ+ T cells isolated from spleen
D: TCRγ/δ+ T cells isolated from peripheral lymph nodes
E: TCRγ/δ+ T cells isolated from thymus
