Mo-DC Generation Tool Box
Overview
The Mo-DC Generation Tool Boxes comprise reagents for magnetic separation of human monocytes and cell culture media for Mo-DC differentiation and maturation. Two different tool boxes are available that contain reagents for the separation of monocytes by either positive selection using CD14 MicroBeads or by depletion of undesired cells using the Monocyte Isolation Kit II, respectively. The Mo-DC Differentiation Medium and the Mo-DC Maturation Medium contain interleukin-4/GM-CSF and TNF-α, respectively. Mo-DCs can easily be differentiated from monocytes with a lot-to-lot consistent, standardized protocol for effective differentiation of immature and mature Mo-DCs.

Downstream applications
For the isolation of monocytes from PBMCs and subsequent differentiation of immature and mature monocyte-derived dendritic cells in vitro. Common uses for Mo-DCs are vaccine development, antigen-specific T cell stimulation, as well as toxicity and hypersensitivity testings.
Columns
For positive selection: MS, LS, or autoMACS® Columns. For depletion: MS, LS, or autoMACS Columns.

Flow cytometric analysis of in vitro generated monocyte-derived dendritic cells (Mo-DCs) using the Mo-DC Differentiation Inspector. Monocytes isolated with CD14 MicroBeads from human PBMCs were differentiated into Mo-DCs in the presence of GM-CSF and IL-4 for 7 days using the Mo-DC Differentiation Medium. Immature Mo-DCs were differentiated into mature Mo-DCs using the Mo-DC Maturation Medium containing TNF-α. Shown are the expression levels of CD14 (A), CD209 (DC-SIGN) (B), and CD83 (C) as mean fluorescence intensities (MFI) on monocytes, immature Mo-DCs (imMo-DCs), and mature Mo-DCs (mMo-DCs) prepared from 12 donors.
