| Description |
PepTivator® Tyrosinase is a peptide pool that consists mainly of 15-mer peptides with 11–amino acid (aa) overlap. PepTivator peptide pools have been developed for the efficient in vitro stimulation of antigen–specific CD4+ and CD8+ T cells as peptides of 15-aa length with 11-aa overlap represent an optimized solution for stimulating both CD4+ and CD8+ T cells in various applications.1 Quantitative, phenotypical, or functional analysis of tyrosinase-specific T cell immunity can provide important information on the natural course of immune responses. |

Background information Besides gp100/Pmel17 and Melan-A/MART-1, tyrosinase belongs to the melanoma-associated antigens of the group of differentiation antigens. It is expressed on normal melanocytes, but is also present in melanoma. Accordingly, CD4+ and CD8+ T lymphocytes recognizing tyrosinase have been identified in melanoma patients. Therefore, tyrosinase represents a potential target for immunotherapy of melanoma2.
Downstream applications The in vitro stimulation of tyrosinase-specific CD4+ and CD8+ T cells with PepTivator Tyrosinase causes the secretion of effector cytokines and the up-regulation of activation markers, which then allow the detection and isolation of tyrosinase–specific T cells.
