Neural Tissue Dissociation Kits
Description
The Neural Tissue Dissociation Kits have been developed for the gentle and efficient generation of single-cell suspensions from neural tissues, and can be used in combination with the gentleMACS™ Dissociator. In less than an hour, the gentle two-step enzymatic dissociation procedure efficiently yields high numbers of viable cells (figure 1). The enzyme mixes provided in the Neural Tissue Dissociation Kit (P) do not contain trypsin, whereas the enzyme mixes of the Neural Tissue Dissociation Kit (T) do not contain papain. The Neural Tissue Dissociation Kit - Postnatal Neurons has been optimized for the special needs of sensitive postnatal neurons during dissociation. For optional further repetitions of the incubation with Enzyme Mix 2 as described within the data sheet, a supplement for the Neural Tissue Dissociation Kit Enzyme Mix 2 is available. Dissociated cells are immediately ready for further in vitro or in vivo applications, including MACS® Cell Separations.
Applications
Dissociation of neural tissues to single-cell suspensions, e.g., for subsequent cell separations using MACS® Technology or further in vitro and in vivo applications. The kit based on papain is best suited for use with CD271 (LNGFR) MicroBeads and Anti-A2B5 MicroBeads, whereas the kit based on trypsin is best suited for use with Anti-PSA-NCAM MicroBeads. CD11b (Microglia) MicroBeads are suitable for use with both kits. The papain-based kit is recommended for use with all other epitopes.

The table shows the recommended kit regarding antigen compatibility for subsequent MACS® Cell Separation. For epitopes not yet
specified in the table, the initial use of the Neural Tissue Dissociation Kit (P) is recommended. Epitope sensitivities have been tested for antibodies available from Miltenyi Biotec.
