The TNF-α Secretion Assay - Cell Enrichment and Detection Kit was developed for the sensitive detection as well as the enrichment of human TNF-α-secreting cells.

Background information
Tumor necrosis factor α (TNF-α) is secreted by activated T cells, monocytes, macrophages, NK cells, and neutrophils. TNF-α is a mediator of inflammatory immune responses and is toxic for many transformed cells, especially in combination with IFN-γ.

Downstream applications
The TNF-α Secretion Assay has been developed for detection and isolation of antigen-specific TNF-α-secreting T cells after stimulation with antigen.
The TNF-α Secretion Assay can also be used to analyze secretion of TNF-α by other leukocytes, e.g., monocytes.
The TNF-α Secretion Assay was used in combination with the IFN-γ Secretion Assay to investigate cytotoxic effects of HIV-specific T cells.1

PBMCs of a CMV+ donor were stimulated for 4 hours with CMV lysate. The responding cells were stained and isolated according to secretion of TNF-α using the TNF-α Secretion Assay - Cell Enrichment and Detection Kit.
In the stimulated sample, 9390 TNF-α-secreting CD4+ T cells were enriched per 106 CD4+ T cells by using MS Columns and a MiniMACS™ Separator. In the unstimulated control sample, no TNF-α-secreting CD4+ T cells were detected per 106 CD4+ T cells after enrichment.
