| IL-10 Secretion Assay - Cell Enrichment and Detection Kit |
|
| Description |
| The IL-10 Secretion Assay - Cell Enrichment and Detection Kit was developed for the sensitive detection as well as the enrichment of human IL-10-secreting cells. |
| Details |
Background information Interleukin-10 (IL-10) is predominantly secreted by human CD4+ T cells and antigen-presenting cells, such as monocytes/macrophages and dendritic cells. IL-10 has important suppressive functions on immune responses and is believed to be involved in the maintenance of tolerance. IL-10 blocks activation of cytokine synthesis by TH1 cells, activated monocytes, and NK cells. It can stimulate the immunoglobulin production by B cells.
Downstream applications IL-10 Secretion Assays were used to analyze IL-10-secreting, CMV-specific CD4+ T cells after stimulation with CMV lysate. The IL-10 Secretion Assay was also used to analyze IL-10 secretion by allergen-specific T cells.1,2 Furthermore, it was used for the detection and isolation of antigen-specific IL-10-secreting type I regulatory T cells from patients with an autoimmune skin disease. The isolated cells were cloned, expanded and further analyzed for their regulatory function.3 |

| Figure 1 |
PBMCs of a CMV+ donor were stimulated for 16 hours with CMV lysate. The responding cells were stained and isolated according to secretion of IL-10 using the IL-10 Secretion Assay - Cell Enrichment and Detection Kit. In the stimulated sample, 329 IL-10-secreting CD4+ cells were enriched per 106 CD4+ T cells by using MS Columns and a MiniMACS™ Separator. In the unstimulated control sample, two IL-10-secreting CD4+ T cells were detected per 106 CD4+ T cells after enrichment. |

| PBMCs of a CMV+ donor were stimulated with CMV-lysate. The responding cells were stained for coexpression of cytokines using the IL-10 Secretion Assay - Detection Kit (PE) and IL-2 Secretion Assay - Detection Kit (APC). The plots show IL-10 and IL-2 secretion by viable CD4+ T cells. |
