Background information Interleukin-4 (IL-4) is predominantly secreted by activated CD4+ memory and effector TH2 cells, basophils, and mast cells. It induces and supports humoral immune responses for the neutralization of extracellular pathogens. The T2 types of immune mechanisms can also be involved in immunologic disorders such as IgE-mediated allergy.
Downstream applications IL-4 Secretion Assay was used to isolate and analyze Tetanus Toxoid-specific1, allergen-specific2,5, and autoreactive7 T cells from PBMCs, as well as tumor-specific cells from T cell lines3. The IL-4 Secretion Assay was also used for isolation of IL-4-secreting human NK cells4, and for detection of IL-4-secreting NKT cells6. IL-4-secreting CD8+ T cells were isolated in combination with CD8 MultiSort MicroBeads.8 |

| PBMCs from a CMV+ donor were stimulated for 16 hours with CMV lysate. The responding cells were stained and isolated according to secretion of IL-4 using the IL-4 Secretion Assay - Cell Enrichment and Detection Kit. In the stimulated sample, 24 IL-4 secreting CD4+ T cells were enriched per 106 CD4+ T cells using MS Columns and a MiniMACS™ Separator. In the unstimulated control sample, two IL-4 secreting CD4+ T cells were detected per 106 CD4+ T cells after enrichment. There seemed to be no difference between stimulated and unstimulated sample before enrichment. |
