| Description |
The CD154/IL-4/CD4 Detection Kit is designed for the intracellular detection of IL-4 production in activated CD4+ T cells. It contains an antibody recognizing CD154 – a specific marker for T cell activation – and all reagents necessary for the immunofluorescent detection of IL-4-producing activated CD4+ T cells from whole blood or PBMCs. By combining the detection of cytokine production with staining of CD154 and CD4, the sensitive and specific detection of activated antigen-specific CD4+ T cells is easily possible. IL-4 is predominantly secreted by activated CD4+ and CD4+ effector and memory T cells, basophils, and mast cells. IL-4 especially induces and supports humoral responses, e.g., by its effects on the activation, proliferation, and antibody production of B cells. |
| Applications |
| The kit is suited for flow cytometric identification and enumeration of IL-4-producing CD154+CD4+ T cells upon in vitro stimulation. Other applications include identification and enumeration of IL-4-producing, antigen-specific CD154+CD4+ T cells upon restimulation or monitoring of specificity of antigen-specific T cell lines. |

| Human PBMCs were incubated with or without CMV pp65 protein for 6 hours. After 2 hours, brefeldin A was added. Cells were fixed, permeabilized, and intracellularly stained with CD154-APC, Anti- IL-4-PE, and CD4-FITC. Cells were analyzed by flow cytometry. |
